Abstract
The perspective of the cytoskeleton as a unique feature of eukaryotic organisms was overturned when homologs of the eukaryotic cytoskeletal elements were identified in prokaryotes, and implicated in major cell functions, including growth, morphogenesis, cell division, DNA partitioning and cell motility.

FtsZ and MreB were the first identified homologs of tubulin and actin, respectively, followed by the discovery of crescentin as intermediate filament-like protein.

In addition, new elements were identified which have no apparent eukaryotic counterparts, such as the deviant Walker A-type ATPases, bactofilins and several novel elements recently identified in streptomycetes, highlighting the unsuspected complexity of cytostructural components in bacteria. In vivo multi-dimensional fluorescence microscopy has demonstrated the dynamics of the bacterial intracellular world and yet we are only starting to understand the role of cytoskeletal elements. Elucidating structure-function relationships remains challenging, because core cytoskeletal protein motifs show remarkable plasticity, with one element often performing various functions and one function being performed by several types of elements.

Structural imaging techniques, such as cryo-electron tomography in combination with advanced light microscopy, are providing the missing links and enable scientists to answer many outstanding questions regarding prokaryotic cellular architecture.

Conferencia de Asilomar (1975)

En una reunión celebrada en California del 24 al 27 de febrero de 1975, un grupo internacional de científicos decidió que debería establecerse un control estricto sobre el uso de la tecnología del DNA recombinante. Esta declaración redactada por el Comité Organizador de la conferencia es el resumen de un informe sometido a la Asamblea de las Ciencias de la Vida de la Academia Nacional de Ciencias y aprobado por su comité ejecutivo el 20 de mayo de 1975.

Recombinant DNA Advisory Committee

The NIH established the Recombinant DNA Advisory Committee (RAC) on October 7, 1974 in response to public concerns regarding the safety of manipulating genetic material through the use of recombinant DNA techniques. Although the RAC's membership and responsibilities have evolved over time with scientific understanding and developments in this technology, it continues to serve the NIH, as well as the scientific and lay publics, as a critically important forum for open, public deliberation on the panoply of scientific, ethical, and legal issues raised by recombinant DNA technology and its basic and clinical research applications. Over the course of the Committee's existence, transparency and access have been its defining characteristics, enabling public acceptance of a critically important technology and creating an environment in which science can advance in an informed, safe, and ethical manner.

La toxina y su antídoto en Streptomyces

Los sistemas toxina-antitoxina de bacterias suelen estar compuestos por un par de genes contiguos que actúan de manera conjunta: uno de ellos codifica una proteína con efecto tóxico mientras que el otro gen codifica la correspondiente proteína antídoto que bloquea la acción de la toxina. Estos sistemas se clasifican en tres tipos, según el mecanismo de acción de la antitoxina. Los más frecuentes son los de tipo II, en los que la toxina es inactivada por su unión a la antitoxina. Las toxinas suelen ser muy resistentes a las proteasas, mientras que las antitoxinas son más sensibles y tienen una vida media mucho más corta. Su función no está muy clara, aunque se han relacionado con la protección contra el ADN extraño, la respuesta al estrés o la muerte celular programada. Estos sistemas han sido muy estudiados en bacterias Gram negativas como Escherichia coli en la que se han identificado al menos 33 sistemas toxina-antitoxina. Suelen estar codificados en plásmidos, por lo que se transmiten entre la población bacteriana con facilidad.

El Instituto de Biología Funcional y Genómica de la Universidad de Salamanca, acaba de publicar en PLoS ONE la primera demostración experimental de uno de estos sistemas toxina-antitoxina funcional en dos especies de Streptomyces. El sistema, similar a uno de E. coli, está compuesto por una proteína YefM que actúa como antitoxina inestable, y por YoeB, que es la toxina estable. La sobre-expresión del sistema YefM/YoeB es letal tanto para E. coli como para de Streptomyces, lo que demuestra que el sistema es funcional. Además el complejo proteico YefM/YoeB purificado interacciona y se une específicamente a determinadas secuencias promotoras, inhibiendo el inicio de la traducción.
Recientemente, mediante análisis bioinformáticos de genomas completos, se han detectado hasta 24 sistemas toxina-antitoxina en Streptomyces, aunque su funcionalidad aún no ha sido demostrada.

Using the poly(dA-dT) “connector” method , a population of annealed hybrid circular DNAs was constructed in vitro; each hybrid DNA circle contained one molecule of poly(dT)-tailed CoI EI-DNA (LRI) annealed to any one of a collection of poly(dA)-tailed linear DNA fragments, produced originally by shearing total E. coli DNA to an average size of 8.5 × 106 daltons. This annealed DNA preparation (12 μg) was used to transform an F+recA E. coli strain (JA200), selecting transformants by their resistance to collcin EI. A collector or “bank” of over 2000 colicin EI-resistant clones was thereby obtained, 70% of which were shown to contain hybrid CoI EI-DNA (E. coli) plasmids. This colony bank is large enough to include hybrid plasmids representative of the entire E. coli genome. Individual plasmids have been readily identified by replica mating the collection onto plates seeded with cultures of various F auxotrophic recipients, selecting for complementation of the auxotrophic markers by F-mediated transfer of hybrid plasmids to the F recipients. In this manner, over 80 hybrid CoI EI-DNA (E. coli) plasmid-bearing clones have been identified in the colony bank, and about 40 known E. coli genes have been tentatively assigned to these various plasmids. The hybrid plasmids are transferred efficiently from F donors to appropriate F recipients. The use of this method to establish similar colony banks in E. coli containing hybrid plasmids representative of various simple eucaryotic genomes is discussed.

PRINCIPIOS DEL MECANISMO TOXINA-ANTITOXINA

La mayoría de las cepas bacterianas portan plásmidos que se mantienen con una cierta estabilidad, pero muchos  plásmidos codifican para sistemas que actúan cuando otros mecanismos de control han fallado, es decir, cuando una bacteria hija no recibe plásmidos después del proceso de división celular.  

Los mecanismos que controlan el mantenimiento de plásmido con genes para  Toxina /Antitoxina son bien conocidos: las antoitoxinas que neutralizan las toxinas son metabólicamente inestables. La rápida disminución de las antitoxinas se produce en células hijas libres de plásmido.  Como dichas células sin plásmido han heredado moléculas de toxina estable de la célula madre, ésta ya no será neutralizada por la antitoxina, y  conduce a la destrucción de las células libres de plásmido. Este mecanismo reduce eficazmente la proliferación de células sin plásmido en  las poblaciones bacterianas.

Abstract

The glyoxylate cycle is an anaplerotic pathway of the tricarboxylic acid (TCA) cycle that allows growth on C(2) compounds by bypassing the CO(2)-generating steps of the TCA cycle. The unique enzymes of this route are isocitrate lyase (ICL) and malate synthase (MS). ICL cleaves isocitrate to glyoxylate and succinate, and MS converts glyoxylate and acetyl-CoA to malate. The end products of the bypass can be used for gluconeogenesis and other biosynthetic processes. The glyoxylate cycle occurs in Eukarya, Bacteria and Archaea. Recent studies of ICL- and MS-deficient strains as well as proteomic and transcriptional analyses show that these enzymes are often important in human, animal and plant pathogenesis. These studies have extended our understanding of the metabolic pathways essential for the survival of pathogens inside the host and provide a more complete picture of the physiology of pathogenic micro-organisms. Hopefully, the recent knowledge generated about the role of the glyoxylate cycle in virulence can be used for the development of new vaccines, or specific inhibitors to combat bacterial and fungal diseases.
